Researchers in Australia have investigated the effects of magnesium L-threonate supplementation on cognitive performance and sleep quality. Their findings indicate that while cognitive enhancements were expected, the significant increase in heart rate variability (HRV) is a novel outcome for magnesium-based products.

Magnesium is a crucial micronutrient required for various functions in the human body, including energy metabolism and muscle contraction. Insufficient magnesium levels are correlated with several health issues such as depression, anxiety, type 2 diabetes, and other cardiometabolic conditions. Alarmingly, about 31% of the global population has inadequate magnesium intake, with this figure rising to 45% in the United States.
The study specifically evaluated magnesium L-threonate, branded as Magtein by Threotech and AIDP, which is noted for its superior bioavailability compared to other magnesium forms. The clinical trial revealed that this form of magnesium improves cognition, reaction time, and memory. Additionally, participants exhibited reduced heart rate and increased HRV, indicating lower stress levels. However, while self-reported improvements in sleep were noted, no significant changes were observed in objective sleep measurements.
“While earlier studies have demonstrated Magtein supports cognitive performance in older populations, this research suggests benefits may also extend to younger adults,” stated Jennifer Gu, Ph.D., senior vice president of research and development for Magtein.
The trial utilized a randomized, double-blind, placebo-controlled design with 100 healthy adults aged 18 to 45 who reported sleep dissatisfaction over six weeks. Participants were given a daily dosage of 2 grams of magnesium L-threonate, divided into two capsules.
Results from the study showed new cognitive improvements, with supplementation correlating to statistically significant increases in cognitive scores, suggesting a decrease in cognitive age by approximately 7.5 years compared to the placebo group. However, the magnesium supplement did not result in notable differences in subjective measures of sleep disturbance, although there was a decrease in sleep impairment scores. Objective sleep data from Oura rings showed a modest reduction in heart rate while sleeping and an increase in HRV among those taking magnesium L-threonate, but no notable differences in overall sleep quality.
Prior studies have also pointed to benefits from magnesium L-threonate, including improved memory and attention in adults. However, earlier sleep studies showed conflicting results regarding sleep quality improvements. Data suggests instead that the impact of the magnesium supplement on sleep may vary and needs further investigation.
