The research indicated that pediatric insomnia can severely impact sleep quality, daytime functioning, academic performance, and overall quality of life, particularly for children with neurodevelopmental disorders such as Autism Spectrum Disorder (ASD) and Attention-Deficit/Hyperactivity Disorder (ADHD). There are limited pharmacological options available for treating pediatric insomnia, with no approved medications in the U.S. and only prolonged-release melatonin authorized in certain pediatric cases in Europe.
The Phase 2 trial assessed three dosages of daridorexant—10 mg, 25 mg, and 50 mg—in children aged 10 to under 18 years diagnosed with insomnia disorder, including those with neurodevelopmental disorders. The findings, which extend understanding of orexin signaling in a younger population, will be detailed by Professor Carmen M. Schröder in the "Impactful Clinical Trial Session II" on October 23.
In addition to the pediatric study, there will be presentations on real-world evidence evaluating daridorexant's impact on insomnia treatment outcomes, including sleep quality, daytime functioning, quality of life, and work productivity.
Interim results from the ProDiGyA study in Germany and Austria will reveal how daridorexant affects real-world treatment patterns and insomnia severity during the initial months of treatment. Results from a Canadian noninterventional study will focus on the drug's influence on quality of life and work productivity for insomnia patients.
Moreover, a new analysis from a pivotal Phase 3 study will evaluate the drug's efficacy and safety concerning short sleep duration in adults. Evaluations across six countries will investigate how the introduction of dual orexin receptor antagonists has modified the usage of traditional sedative-hypnotics and the associated healthcare policies affecting their adoption.
At least six investigator-initiated studies on daridorexant will also be presented, addressing its effects on various populations, including menopausal women and patients with epilepsy or major depressive disorder. These collective efforts aim to enhance understanding of insomnia management and contribute to the evolving landscape of treatment options.
