Sleep is crucial for overall health and emotional well-being, and the amounts and quality of sleep can significantly affect physiological functions. This new research expands on prior findings that low sleep correlates with faster brain aging. The study, conducted by a team led by Junhao Wen, PhD, from Columbia University, examined biological aging across multiple organs in relation to sleep duration. Wen emphasized that sleep is vital for healthy aging and could be modified for better health outcomes.

Biological age refers to the rate at which cells and tissues age, rather than the chronological age based on years lived. The research team used data from approximately half a million participants in the UK Biobank and machine learning techniques to analyze aging signatures across various organs. They focused on self-reported sleep duration, using 23 aging clocks spanning 17 organs, with the study defining insufficient sleep as less than 6 hours and excessive sleep as more than 8 hours. Participants sleeping between 6.4 and 7.8 hours per night did not show the same signs of accelerated biological aging as those sleeping too little or too much.

While the findings indicate a relationship between sleep duration and biological aging, they do not imply direct causation. However, both inadequate and excessive sleep were linked to poorer overall health. Sarathi Bhattacharyya, MD, a specialist in sleep medicine, noted that short sleep durations correlate with immune system issues, increased inflammation, disrupted metabolic processes, and heightened sympathetic nervous system activity—all factors that may contribute to faster cellular aging.

Conversely, prolonged sleep may reflect underlying health issues that can also accelerate aging. The study also established links between inadequate sleep and a variety of chronic conditions, including obesity, type 2 diabetes, hypertension, ischemic heart disease, and heart arrhythmias. Alex Dimitriu, MD, highlighted that insufficient sleep often relates to the stress of busy lifestyles and may raise blood pressure and glucose levels, with long sleep often associated with illness rather than a simple recovery.

The research utilized organ-specific aging clocks to delve deeper into the connection between sleep and late-life depression. While the study could not definitively clarify causal relationships between sleep duration and depression, mediation analysis suggested that insufficient sleep may directly contribute to the burden of late-life depression, whereas excessive sleep might be related through underlying health factors.

Wen noted that the study’s outcomes could influence future research and clinical approaches to sleep management. He advocated for personalized sleep optimization strategies considering individual medical histories, suggesting that short and long sleepers need different interventions. Experts recommend a sleep window of at least eight hours to achieve seven quality hours of rest, emphasizing good sleep hygiene and consistent wake times as essential components for maintaining a healthy circadian rhythm.
